Principle of Bacterial Protein Expression

Bacterial expression produces recombinant proteins by introducing a plasmid carrying the gene of interest under a regulated promoter into a prokaryotic host such as E. coli. Upon induction, the bacterial transcription and translation machinery synthesizes the target protein, which may remain in the cytoplasm, be directed to the periplasm through signal peptides, or accumulate as inclusion bodies depending on sequence properties and expression conditions. While bacterial systems do not perform most eukaryotic post-translational modifications, they are widely used for enzymes, antigens, and non-glycosylated proteins where speed, yield, and cost-efficiency are primary constraints.
