Protein Expression Technical Process

Nebulabio’s production process is a systematic, stage-gated workflow, optimized for each host system to maximize the probability of delivering a functional protein.

 Phase 1: Construct Design & Vector Engineering 

  • Gene Optimization: Your target gene sequence is analyzed and codon-optimized for the selected host organism to maximize translation efficiency and protein yield.

  • Vector Assembly: The gene is cloned into an expression vector optimized for the host. Key elements include:

  1. Promoter/Inducer: (e.g., T7/lac for E. coli, polyhedrin/p10 for baculovirus, CMV for mammalian).

  2. Fusion Tags: Strategic addition of tags (His-tag, GST, MBP, SUMO, Fc) to enhance solubility, enable purification, or improve detection.

  3. Secretion Signals: For systems like yeast or mammalian cells, signals are added to direct the protein to the culture supernatant for easier purification.


 Phase 2: Host Cell Transformation & Screening 

  • Small-Scale Expression Screening: Multiple constructs (e.g., with different tags, truncations, or vector backbones) are tested in parallel at a small scale (mL culture).

  • Condition Optimization: We test variables like temperature, inducer concentration, induction time, and media composition to find the conditions that maximize soluble yield.

  • Analytical Assessment: Small-scale cultures are analyzed by SDS-PAGE and Western blot to identify the most promising expression clone and conditions.


 Phase 3: Scale-Up Production 

  • Process Scale-Up: The optimized protocol is scaled to the required volume—from shake flasks (100 mL - 1 L) to bioreactors (1 L - 100+ L)—with careful monitoring and control of pH, dissolved oxygen, and nutrient feeding to maintain cell health and protein productivity.

  • Harvest: Cells are separated from the culture medium via centrifugation or filtration. For intracellular proteins, cells are lysed using mechanical (sonication, homogenization) or chemical methods.


 Phase 4: Protein Purification & Refolding 

  • Capture: The target protein is isolated using affinity chromatography corresponding to its tag (e.g., IMAC for His-tag, Glutathione resin for GST-tag, Protein A for Fc-fusion).

  • Tag Cleavage (Optional): If a cleavable tag (e.g., TEV, HRV 3C protease site) was used, it is removed at this stage.

  • Polishing: Further purification steps—such as ion exchange chromatography (IEX), hydrophobic interaction chromatography (HIC), or size exclusion chromatography (SEC)—are employed to achieve high purity, remove aggregates, contaminants, and cleaved tags.

  • Refolding (if needed): For proteins expressed in inclusion bodies (E. coli), we have specialized protocols for solubilizing the denatured protein and refolding it into its native, active conformation.


 Phase 5: Characterization & Quality Control 

  • Purity: Assessed by SDS-PAGE (Coomassie/silver stain) and analytical SEC-HPLC.

  • Identity: Confirmed by mass spectrometry (MALDI-TOF or LC-MS).

  • Concentration: Precisely determined by spectrophotometry (A280) or BCA assay.

  • Activity & Functionality: Validated using client-specified functional assays (e.g., enzymatic activity, ligand binding by ELISA/SPR/BLI, cell-based assays).

  • Biophysical Properties: Analyzed for homogeneity, thermal stability (DSF), and endotoxin levels as required.
