Custom Gene Expression Cell LinesAbout Us

Nebulabio is a cell engineering team specializing in mammalian stable cell line development for research and discovery workflows. By combining standardized project management with flexible technical options, we deliver assay-ready cell lines with clear documentation, traceable milestones, and practical QC. Our goal is simple: you provide the gene-of-interest (GOI) and intended application, and we deliver a stable expression system you can trust and reproduce.

Nebulabio’s Stable Cell Line Platform

Stable cell line development is a robust mammalian cell engineering workflow that enables long-term, consistent expression of your gene-of-interest by integrating and selecting expression cassettes in the host genome, providing assay-ready models with improved reproducibility across passages. Our comprehensive platform covers construct strategy and gene delivery, stable pool generation with optional single-cell cloning, and stability testing with QC and cell banking to accelerate your functional studies and screening programs.

Supported Expression Options

  • Host Cell Lines (HEK293 / CHO / Customer-Provided Lines):

Supports widely used mammalian hosts for robust protein expression and assay development; customer-supplied or disease-relevant cell lines are also accepted upon feasibility evaluation.

  • Constitutive Expression Systems:

Enables continuous, stable expression of the gene-of-interest (GOI) across passages—ideal for routine functional assays, screening, and long-term phenotyping.

  • Inducible Expression Systems (Optional):

Provides tightly controlled, on-demand expression (e.g., for toxic or growth-inhibitory genes), improving cell fitness and experimental flexibility.

  • Gene Delivery Strategies (Transfection / Electroporation / Lentiviral Transduction):

Multiple delivery routes are available to match cell type, efficiency requirements, and project timelines, ensuring reliable integration and expression.

  • Stable Pools (Polyclonal) vs. Single-Cell Derived Clones:

Stable pools offer faster turnaround and cost efficiency; monoclonal lines provide more uniform expression and higher assay stringency for demanding applications.

  • Detection & Validation Readouts (qPCR / Western Blot / Flow / ELISA / Functional Assays):

Expression confirmation is tailored to your target type and use case, covering transcriptional, protein-level, surface expression, secretion, and functional endpoints.

Principle of Stable Gene Expression

Stable gene expression enables the generation of mammalian cell lines that maintain consistent, long-term production of a gene-of-interest (GOI) by introducing an expression cassette into host cells and enriching for cells that stably retain and express the transgene. After gene delivery (e.g., transfection, electroporation, or viral transduction), selective pressure is applied to establish a stable population, and expression-positive cells are expanded to form a stable pool; when higher uniformity is required, single-cell cloning further isolates cell lines with robust and consistent expression. As a result, cells exhibiting the desired expression level and performance (e.g., protein abundance, surface display, secretion, or functional activity) are enriched and stabilized across passages.

Compared with transient expression, stable expression provides assay-ready models with improved reproducibility and reduced batch-to-batch variability, which is critical for functional assays, screening, and long-term phenotyping. By combining a structured selection process with fit-for-purpose validation, stable expression workflows deliver reliable cellular tools that support mechanistic studies and downstream assay development, rather than short-lived, variable expression outcomes typically observed in transient systems.

Custom Gene Expression Cell Lines

 1. Project Design & Input Preparation

We translate your gene-of-interest (GOI) and application needs into a buildable, assay-oriented stable expression strategy.

Project Scoping:

  • Confirm GOI (species/isoform), expression mode (constitutive or inducible),

tag (optional), host cell line (HEK293/CHO/customer-provided), and intended assay readouts.

Input Options:

  • Provide GOI sequence / gene name, or

  • Provide your plasmid/expression cassette.

Recommended Output Tier:

  • Stable pool (polyclonal) for speed and cost-efficiency, or

  • Single-cell derived clone(s) for uniformity and high-stringency assays.

 2. Vector Construction & Virus Packaging

We construct and QC the expression vector, and generate high-quality delivery material based on the chosen strategy.

Key Steps:

  • Vector design: promoter/marker/tag strategy aligned to your cell type and endpoint

  • Cloning & sequence confirmation: Sanger-confirmed insert and correct reading frame (when applicable)

  • Plasmid preparation: transfection-grade DNA

  • Optional: lentivirus packaging: high-quality viral preparation for hard-to-transfect cells

QC & Documentation:

  • Plasmid map + sequence confirmation record

  • Delivery material QC summary (project-dependent)

 3. Gene Delivery & Stable Pool Establishment

We introduce the GOI into host cells and establish a stable expressing population under selection.

Key Steps:

  • Gene delivery options: plasmid transfection / electroporation / lentiviral transduction

  • Selection & expansion: antibiotic selection → recovery → stable pool expansion

  • Early expression check: quick confirmation that expression signal is present before deep screening

Common methods (as applicable):

  • Drug selection

  • FACS enrichment (optional, for fluorescent tags/reporters or surface targets)

  • Sub-culturing & stabilization

 4. Single-Cell Clone Generation (Optional)

When monoclonality and expression uniformity are required, we isolate and screen single-cell derived clones.

Key Steps:

  • Single-cell isolation: limiting dilution or single-cell sorting

  • Clonal expansion: recover and scale-up

  • Primary clone screening: expression level + growth performance

What you get (clone tier):

  • A shortlist of top-performing clones (number configurable)

  • Screening summary table (expression vs. growth)

 5. Validation, Genotype Confirmation & QC

We verify expression using fit-for-purpose assays and complete QC to support reproducibility.

Core validation options (select per target type):

  • RT-qPCR

  • Western blot (WB) / ELISA

  • Flow cytometry

  • Immunofluorescence (IF)

  • Functional assays (pathway/response readouts; project-defined)

Genotype / construct confirmation (as needed):

  • PCR-based confirmation (project-dependent)

  • Sanger sequencing of junctions/insert (if applicable)

QC (recommended):

  • Mycoplasma test (standard)

  • Sterility/contamination checks (as applicable)

 6.Cell Banking & Delivery

We cryopreserve and ship the final stable pool or clone(s) with a complete documentation package.

Deliverables (industry-standard):

  • Cryopreserved stable cells (multiple vials)

  • COA / QC summary (mycoplasma status + configured QC items)

  • Validation data package (assay results used for acceptance)

  • Culture & recovery guidance (medium, passaging notes, recommended passage window)
