About Us

未命名.pngNebulabio is a cell engineering team specializing in mammalian stable cell line development for research and discovery workflows. By integrating standardized project execution with flexible genome editing strategies, we deliver assay-ready knockout cell lines with clear documentation, traceable milestones, and practical quality control—enabling consistent, reproducible experiments that support confident interpretation of loss-of-function phenotypes.

Our Stable Cell Line Platform

Our platform integrates project design and feasibility assessment, construct or genome editing strategy selection, gene delivery and enrichment, stable pool establishment with optional single-cell cloning, and systematic validation, quality control, and cell banking. The resulting cell lines are delivered as ready-to-use research tools aligned with defined downstream applications rather than generic engineered cells.

Supported Stable Cell Line Services

■ Stable Expression Cell Lines

  • Constitutive or inducible expression of a defined gene of interest

  • Suitable for protein expression, functional studies, and assay development

■ Reporter Cell Line Generation

  • Pathway-responsive, promoter-driven, or endogenous reporters

  • Luciferase, fluorescent, or secreted reporter formats

  • Designed for quantitative readouts in screening and mechanism-of-action studies

■ Gene Knockdown Cell Lines

  • Stable RNAi systems (shRNA / miR-shRNA) with optional CRISPRi approaches

  • Designed for durable partial loss-of-function and pathway interrogation

■ Knock-in Cell Line Generation

  • Precise insertion at endogenous loci or safe harbor sites

  • Supports reporters, epitope or fluorescent tags, functional cassettes, and defined variants

■ Knockout Cell Line Generation

  • CRISPR-based gene disruption (indel knockout, deletion knockout)

  • Optional multiplex or conditional knockout strategies

  • Designed for permanent loss-of-function models with strong genetic causality

■ Delivery Methods (Transfection / Electroporation / Lentiviral Transduction)

Flexible delivery routes are selected based on host cell compatibility, efficiency requirements, and project timelines.

■ Stable Pools (Polyclonal) vs. Single-Cell–Derived Clones

  • Stable pools enable faster turnaround and cost efficiency for early-stage studies

  • Monoclonal lines provide uniform behavior and defined genotype for high-stringency workflows

■ Validation and QC Options (Fit-for-Purpose)

  • DNA-level confirmation (PCR, Sanger sequencing; junction checks for knock-in)

  • Expression or phenotype confirmation (qPCR, Western blot, flow cytometry, imaging, luminescence)

  • Standard quality control (mycoplasma testing and contamination checks)

  • Optional assay robustness metrics (CV, dynamic range, Z’ factor for screening applications)

Principle of Stable Cell Lines

Stable cell lines are generated by introducing a defined genetic modification—such as stable expression, reporter integration, knockdown, knock-in, or knockout—and enriching for cells that retain the modification over long-term culture. Following gene delivery or genome editing, cells are expanded as a stable pooled population under defined selection or enrichment conditions.

When experimental requirements demand uniformity and genotype certainty, single-cell cloning is used to isolate individual clones that are screened for correct genotype, performance characteristics, and growth behavior. Compared with transient systems, stable cell lines provide consistent outputs across passages and experimental batches, enabling reliable downstream applications.

Nebulabio Service Workflow

Stable Cell Line Services

 1. Project Design & Input Preparation

We translate your biological objective into a buildable, assay-oriented stable cell line strategy.

Project Scoping:

  • Modification type: stable expression, reporter, knockdown, knock-in, or knockout

  • Host cell line selection and feasibility assessment

  • Control design (parental, vector control, non-target control, rescue plan when relevant)

  • Acceptance criteria aligned with downstream assays (expression level, fold induction, knockdown percentage, genotype confirmation, etc.)

Inputs You Can Provide:

  • Target gene or genomic locus and species information

  • Insert sequences, promoters, or response elements (as applicable)

  • Preferred vectors or plasmids (optional)

  • Intended assay conditions and readouts

 2. Construct Design, Reagent Preparation & Delivery Planning

We prepare the required vectors or genome editing reagents and define the most reliable delivery strategy.

Key Steps:

  • Vector design and cloning (expression, reporter, shRNA)

  • Guide RNA and donor design (CRISPR knockout or knock-in)

  • Sequence confirmation (as applicable)

  • Delivery route selection: transfection, electroporation, or lentiviral transduction

  • Selection and enrichment planning (antibiotics, fluorescence-based sorting, etc.)

 3. Gene Delivery & Stable Pool Establishment

We generate and expand stable genetically modified cell populations.

Key Steps:

  • Delivery of constructs or genome editing reagents

  • Application of selection or enrichment and recovery

  • Expansion of stable pools

  • Early checkpoint assays to confirm modification before proceeding to cloning

 4. Single-Cell Clone Generation (Optional)

For applications requiring defined genotype and high uniformity, we isolate and evaluate monoclonal lines.

Key Steps:

  • Single-cell isolation by sorting or limiting dilution

  • Clonal expansion

  • Primary screening for genotype and/or functional performance

  • Clone ranking and shortlist selection

 5. Validation, Genotype Confirmation & QC

We confirm that the cell line meets defined performance and quality criteria.

Common Validation Modules (Selected per Project):

  • Expression: qPCR, Western blot, flow cytometry, ELISA

  • Reporter performance: baseline signal, inducibility, dynamic range, dose response
