Principle of Stable GPCR Cell Lines

Stable GPCR cell lines are generated by introducing a GPCR expression cassette—optionally combined with pathway or accessory components—into host cells and enriching for populations that maintain receptor expression and functional signaling over long-term culture. Following gene delivery, selective pressure establishes a stable pool, and receptor-positive cells are expanded under conditions that preserve surface expression and signaling competence.


When assay stringency requires greater uniformity, single-cell cloning isolates monoclonal lines with optimized expression, low background, robust ligand responsiveness, and stable pharmacology across passages. This approach produces GPCR models that function as reproducible assay systems rather than transient expression readouts.

 1. Project Design & Input Preparation 

We translate your GPCR pharmacology objective into a buildable, assay-oriented stable cell line strategy.

Project Scoping:

  • GPCR target (species/isoform) and expression format (native vs tagged)

  • Host cell line selection (HEK293 / CHO / customer-provided)

  • Intended signaling pathway and assay readout

  • Accessory component requirements (G proteins, RAMPs, arrestin modules)

  • Acceptance criteria (baseline, fold response, EC50/IC50 window, CV, Z’ if screening)

Input Options:

 2. Vector Construction & Accessory Module Preparation 

We construct and QC GPCR expression vectors optimized for stable signaling.
