Overview
Nebulabio offer a sophisticated Mouse Immunized Antibody Library construction service, a powerful platform for accessing a rich, high-affinity, and functionally diverse repertoire of antibodies derived from the natural murine immune response. This service is designed to empower therapeutic antibody discovery, diagnostic reagent development, and high-value target validation. By immunizing mice with your target antigen (recombinant proteins, peptides, cells, or pathogens), we harness the full power of in vivo affinity maturation and selection to generate B cells producing antibodies with superior specificity and biological relevance. Our service goes beyond simple library construction; we provide the complete pipeline from antigen design and animal immunization to the generation of ready-to-screen phage display or yeast display libraries, or the direct recovery of paired VH:VL sequences via NGS. This platform is ideal for discovering lead candidates against complex targets where conformational epitopes, post-translational modifications, or functional neutralization are critical.
☞Introduction to Mouse Immunized Antibody Library
A Mouse Immunized Antibody Library is a collection of antibody gene sequences (typically as Fab or scFv fragments) cloned from the B lymphocytes of immunized mice. It captures the immune system's natural response to the antigen, offering distinct advantages over naïve or synthetic libraries:
High Affinity & Matured Sequences: The in vivo immune response involves somatic hypermutation and clonal selection, resulting in antibody sequences that are pre-optimized for high-affinity binding to the target. This yields a higher proportion of potent binders compared to naïve libraries.
Functional & Conformational Epitope Bias: Immunization with properly folded antigens, especially whole cells or membrane proteins, drives the immune response towards functionally relevant, conformational epitopes. This increases the likelihood of discovering antibodies that can modulate biological activity (agonists/antagonists) or neutralize pathogens.
Diversity from a Proven System: The mouse immune system is exceptionally well-characterized. Using defined mouse strains (e.g., BALB/c, C57BL/6) allows for predictable immune responses and compatibility with subsequent hybridoma development if desired.
Compatibility with Display Technologies: The amplified VH and VL genes from immune B cells can be cloned into phage, yeast, or mammalian display vectors. This creates an "immune library" enriched for target-specific clones, significantly reducing the screening effort required to find high-quality hits.
Our service strategically leverages this natural selection process to build focused, high-value libraries primed for success in downstream screening campaigns.

☞Mouse Immunized Antibody Library Development Process
Our development process is meticulously designed to maximize library quality, diversity, and functionality.
①Antigen Design & Immunization Strategy: We collaborate with you to design the optimal immunogen (full-length protein, domain, peptide-carrier conjugate, etc.) and a tailored immunization protocol (adjuvant, schedule, route). Using multiple mice and varied regimens helps capture a broader immune response.
②Immunization & Immune Monitoring: Mice are immunized under strict ethical guidelines. Serum titers are monitored via ELISA to confirm a robust antigen-specific antibody response before proceeding.
③Lymphocyte Harvest: At the peak of the immune response, spleen and/or lymph nodes are harvested. These tissues are rich in antigen-experienced B cells and plasma cells.
④RNA Isolation & cDNA Synthesis: Total RNA is extracted from the lymphocyte pool. This RNA serves as the template for cDNA synthesis, which contains the antibody gene transcripts.
⑤Amplification of Antibody Gene Repertoire: Using family-specific or degenerate primers, the variable regions of the heavy chain (VH) and light chains (Vκ and Vλ) are amplified via PCR. Special attention is paid to maintaining the natural pairing of chains—either by keeping them separate for random recombination in phage display or by using overlap-extension PCR to create linked scFv fragments.
⑥Library Construction & Cloning: The amplified antibody gene fragments are cloned in bulk into the chosen display vector (phagemid for phage display, yeast display vector). This involves ligation or recombination-based cloning to create a diverse library of individual clones, each displaying a unique antibody fragment on its surface.
⑦Library Quality Control & Analysis: The primary library is characterized for:
Diversity: Determined by transforming the ligation product and counting colony-forming units (CFU). A high-quality library typically contains >10^8 independent clones.
Insert Rate: Verified by colony PCR or restriction digest to ensure a high percentage of clones contain the correct insert.
Sequence Diversity: Next-Generation Sequencing (NGS) can be performed on the pooled plasmid library to assess the distribution of V-gene families and overall sequence diversity.
The final deliverable is a titered, ready-to-pan immune antibody library in your chosen display format, accompanied by a full QC report.

Library QC & Analysis: Rigorous quantification and qualification of the library, including titer determination, insert check, and optional deep sequencing analysis to confirm diversity.
Final Delivery: Shipment of the validated library (as purified DNA or as transformed cells, e.g., electroporated E. coli for phage libraries), along with a comprehensive technical report detailing all steps and QC data.

☞Applications of Mouse Immunized Antibody Library
Therapeutic Antibody Discovery: The primary application. The immune library is panned against a therapeutic target (e.g., a cancer cell surface antigen, cytokine receptor) to rapidly isolate high-affinity, functional lead candidates with blocking or neutralizing activity, significantly accelerating early-stage drug discovery.
Diagnostic & Research Reagent Generation: Panning against biomarkers, viral antigens, or specific protein isoforms to develop highly specific capture and detection antibodies for ELISA, lateral flow assays, or immunohistochemistry.
Membrane Protein & Complex Target Targeting: Immunization with whole cells or membrane preparations allows for the generation of antibodies against native, conformationally intact membrane proteins (GPCRs, ion channels), which are often difficult to target with other methods.
Antibody Optimization & Humanization: The murine-derived lead antibodies from the library provide an excellent starting point for subsequent affinity maturation campaigns and humanization for therapeutic development.
Epitope Mapping & Binning: Immune libraries can be used to generate panels of antibodies against different epitopes on the same target, useful for understanding functional domains and for developing non-competing antibody pairs for diagnostic assays.
