Protein Purification Process

Protein purification is the strategic separation of a target protein from a complex mixture of contaminants, including host cell proteins (HCPs), nucleic acids, lipids, endotoxins, viruses, and product-related impurities (aggregates, fragments). Our process follows a proven, orthogonal approach, where multiple purification techniques with different separation mechanisms are combined to achieve the required purity and safety profile.

Our Core Purification Workflow:

 Step 1  Capture & Initial Purification

 Step 2  Intermediate Purification (Polishing)

The primary goal is to isolate and concentrate the target protein from a large volume of crude starting material (e.g., cell culture supernatant, lysate, serum).

  • Affinity Chromatography: The most powerful initial step when available. We utilize specific, reversible biological interactions (e.g., Protein A/G for antibodies, immobilized metal affinity chromatography [IMAC] for His-tagged proteins, ligand-based for enzymes).

  • Ion Exchange Chromatography (IEX): Often used as a first step for non-tagged proteins. Separates proteins based on net charge at a specific pH.


   

This phase removes the bulk of remaining impurities that co-eluted during capture.

  • Ion Exchange Chromatography (IEX): In bind-and-elute or flow-through mode, effectively removes HCPs, DNA, and aggregates based on charge differences.

  • Hydrophobic Interaction Chromatography (HIC): Separates proteins based on surface hydrophobicity. Excellent for removing aggregates and closely related variants.

Quality Control (QC)

The final steps ensure the removal of trace impurities and prepare the protein for its end use.

  • Size Exclusion Chromatography (SEC/Gel Filtration): Separates based on molecular size. This is the gold standard for removing aggregates (high molecular weight, HMW) and fragments (low molecular weight, LMW). It also serves as a gentle method for buffer exchange into the final formulation buffer.

  • Viral Clearance (for therapeutic proteins): Dedicated steps like virus filtration (nanofiltration) and low-pH hold are integrated to meet regulatory safety requirements.

  • Ultrafiltration/Diafiltration (UF/DF): Concentrates the protein to the target concentration and performs final buffer exchange.

  
  • Quality is assessed at every stage. The final product undergoes a comprehensive analytical panel to confirm it meets all specifications.
