Our service leverages a remarkable natural biological principle. Camelids, such as llamas, possess a unique adaptive immune system that generates a significant proportion of heavy-chain-only antibodies (HCAbs) alongside conventional antibodies. These HCAbs lack light chains and the first constant domain (CH1). Their antigen-binding capability is contained within a single variable domain, the VHH (also known as a Nanobody).

☞Llama VHH Antibody Production Process
Our production pipeline is a multi-stage, integrated process designed for quality and yield at every step.
Phase 1: Llama Immunization & Immune Response Monitoring
Project Design & Antigen Preparation: We consult on antigen design (protein, peptide, cell, etc.) and formulation with appropriate adjuvants to maximize immune response diversity and titer.
Ethical Immunization Program: Conducted at our certified animal facility under strict ethical guidelines. A multi-injection schedule over 6-8 weeks ensures a mature, high-affinity immune response.
Serum Monitoring: Regular bleeds are taken to monitor antigen-specific antibody titers via ELISA, ensuring immunization proceeds optimally before the final lymphocyte harvest.
Phase 2: VHH Gene Library Construction & Selection
Lymphocyte Isolation: Peripheral blood lymphocytes (PBLs) are collected from the immunized llama at the peak of the immune response.
Phage Display Library Construction: RNA is extracted, and VHH genes are specifically amplified via RT-PCR using primers targeting the unique features of camelid HCAb mRNA. The genes are cloned into a phage display vector to create an immune VHH library.

Phase 3: Recombinant VHH Expression & Small-Scale Production
Clone Selection & Vector Construction: The DNA sequence of the selected VHH clone(s) is codon-optimized for the chosen expression host (typically E. coli for cytoplasmic or periplasmic expression) and cloned into a high-expression vector (e.g., with a His-tag for purification).
Expression Screening: Small-scale cultures are used to optimize expression conditions (temperature, inducer concentration, time).
Small-Scale Purification: VHHs are expressed and purified via immobilized metal affinity chromatography (IMAC) using the His-tag, followed by size-exclusion chromatography (SEC) or dialysis for buffer exchange and polishing.
Phase 4: Scale-Up Production & Quality Control
Fermentation Scale-Up: The optimal expression protocol is scaled to liter-scale fermentation in bioreactors for higher yield.
Large-Scale Purification: IMAC and SEC are performed at scale to produce milligram to gram quantities of pure VHH.
Comprehensive Quality Control (QC): Every batch undergoes stringent QC:
Purity: >95% as assessed by SDS-PAGE and analytical SEC.
