☞Introduction to Rabbit Derived Antibodies
Rabbit-derived antibodies offer a compelling set of advantages that have solidified their status as a gold standard, particularly for research and diagnostic applications.
Superior Immune Response: Rabbits possess a larger and more diverse immune repertoire compared to mice. They often generate antibodies with higher affinity and against a broader range of epitopes, including those that may be poorly immunogenic in rodents. This makes them ideal for targets where high sensitivity is critical.
Advantages for Polyclonal Antibodies (pAbs): Rabbit polyclonal antisera are exceptionally powerful tools. Their multi-epitope recognition results in strong signals in applications like Western Blotting and Immunohistochemistry (IHC), even for low-abundance or denatured targets. They are also excellent for immunoprecipitation (IP) due to their ability to capture target proteins efficiently from complex lysates.
Advancements in Monoclonal Antibodies (mAbs): While historically more challenging than murine hybridomas, rabbit monoclonal antibody technology has matured significantly. Rabbit mAbs combine the traditional benefits of monoclonality (unlimited supply, consistency) with the inherent advantages of the rabbit immune system: exceptionally high affinity (often in the pico- to nanomolar range) and the ability to recognize unique epitopes not commonly targeted by mouse antibodies. This is especially valuable for distinguishing between closely related protein isoforms or post-translationally modified targets.
Flexible Immunogen Compatibility: Rabbits respond robustly to various immunogens, including small peptides (conjugated to carriers), recombinant proteins (even from human or mouse origin), and complex antigens like glycoproteins or viral particles.
☞Rabbit Derived Antibody Production Process
Nebulabio’s process is tailored to maximize success for both polyclonal and monoclonal outputs, adhering to high ethical and scientific standards.
A. Polyclonal Antibody Production:
Immunogen Design & Conjugation: We assist in selecting optimal antigen sequences (e.g., peptide vs. full-length protein) and perform necessary conjugations (e.g., KLH conjugation for peptides).
Immunization & Boosting: Multiple rabbits are immunized using customized adjuvants and schedules to maximize immune response while ensuring animal welfare. Pre-immune serum is collected as a critical negative control.
Test Bleeds & Titer Monitoring: Serum is drawn at regular intervals to monitor antigen-specific antibody titers via ELISA, allowing us to track immune response progression.
Final Production Bleed: Once the target titer is achieved, a large-volume terminal bleed is performed to collect the final antiserum.
Serum Processing & Optional Purification: Serum is processed (clotted, centrifuged, filtered) and can be provided as whole antiserum or further purified to isolate the total IgG fraction or undergo antigen-affinity purification for enhanced specificity.
B. Monoclonal Antibody Production (Rabbit Hybridoma/Single B Cell Technology):
Immunization & Lymphocyte Isolation: Rabbits are immunized to elicit a potent immune response. Upon confirmation of high titers, spleen cells or peripheral blood lymphocytes are harvested.
Fusion & Hybridoma Generation (Traditional Method): Rabbit B cells are fused with a specialized fusion partner cell line (e.g., a murine myeloma variant) to create immortalized hybridomas.
Single B Cell Sorting & Cloning (Advanced Method): As an alternative, antigen-specific B cells can be isolated via fluorescence-activated cell sorting (FACS) using labeled antigen. Antibody genes are then recovered from single cells by RT-PCR, bypassing the fusion step.
Screening & Subcloning: Regardless of the method, thousands of clones are screened using high-throughput assays (ELISA, FACS) to identify those secreting the desired antibody. Positive clones are subcloned to ensure monoclonality and stability.
Recombinant Expression & Scale-up: The genes for the selected antibody are cloned into expression vectors and produced recombinantly in mammalian cells (e.g., HEK293) for consistent, scalable production, moving away from ascites methods.
Purification & Characterization: Antibodies are purified via Protein A/G affinity chromatography and undergo full characterization, including affinity measurement (SPR/BLI), epitope mapping, and specificity profiling.

☞Nebulabio’s Service Process
Our client-centric workflow ensures clarity, collaboration, and timely delivery of high-quality reagents.

Project Initiation: Collaborative consultation to define the antigen, desired antibody properties (polyclonal vs. monoclonal), and application requirements.
Immunization & Monitoring: Execution of the customized immunization protocol in rabbits, with regular test bleeds and titer analysis to guide the process.
Antibody Generation Path:
Polyclonal Path: Proceeds to final bleed, serum collection, and optional purification.
Monoclonal Path: Diverges to B cell fusion or sorting, followed by intensive screening to isolate a single, high-producing clone.
Production & Purification: For monoclonals, this involves recombinant expression and affinity purification. For polyclonals, it involves serum processing and IgG purification.
Characterization & QC: All final products undergo rigorous testing for specificity, affinity (for mAbs), titer (for pAbs), purity, and functionality in a relevant assay.
Delivery: The purified antibody (and hybridoma cell line if requested) is shipped along with a comprehensive report detailing all protocols and QC data.
☞Advantages and Applications of Rabbit Derived Antibodies
Key Advantages:
High Affinity & Sensitivity: Rabbit antibodies often exhibit 10-100 times higher affinity than their murine counterparts, leading to more sensitive detection in assays.
Broad Epitope Recognition (pAbs): Polyclonal sera recognize multiple epitopes, making them robust reagents for detecting proteins that may be partially denatured or have masked epitopes.
Superior Specificity (mAbs): Rabbit monoclonal antibodies can be generated against epitopes that are non-immunogenic or poorly immunogenic in mice, providing unique specificity tools.
Excellent Performance in Common Assays: They are the preferred choice for demanding applications like Western blotting, IHC, and IP due to their strong signal and high specificity.
Primary Applications:
Biomedical Research: Essential reagents for protein detection, localization, and functional studies across all disciplines.
Diagnostic Development: Widely used as capture and detection antibodies in high-sensitivity ELISA kits, lateral flow assays, and IHC-based companion diagnostics.
Therapeutic Discovery: Rabbit monoclonal antibodies are increasingly used as starting points for humanized therapeutic antibodies due to their high initial affinity and unique epitope targeting.
Cell Biology and Signaling: Critical for studying post-translational modifications (phosphorylation, acetylation) where high-specificity antibodies are required.
