①HT Gene Synthesis & Cloning:
Input: Hundreds to thousands of antibody variable region (VH/VL) sequences, provided by the client or derived from sequencing campaigns (e.g., from single B cell or phage display projects).
Process: Sequences are codon-optimized for mammalian expression. Using automated liquid handlers, genes are synthesized or PCR-amplified in 96- or 384-well plate formats and simultaneously cloned into our standard mammalian expression vectors (e.g., containing a CMV promoter and a constant region for IgG, Fab, or scFv format).
Output: A verified, high-density plate library of antibody expression constructs.
②HT Transfection & Expression:
Platform: We utilize transient transfection in HEK293 cells as our primary HT workhorse for its rapid turnaround (7-10 days) and proven ability to produce correctly folded, glycosylated antibodies. For projects requiring higher titers or specific glycoforms, we offer parallelized stable pool generation in CHO cells.
